Regulated cell death RCD or programmed cell death PCD is not only essential in embryonic development, but also plays an important role in the occurrence and progression of diseases, especially cancers. Escaping of cell death is one of hallmarks of cancer. Gasdermin family proteins are the executors of pyroptosis.
Pyroptosis
Currently, pyroptosis has received more and more attention because of its association with innate immunity and disease. The research scope of pyroptosis has expanded with the discovery of the gasdermin family. A great deal of evidence shows that pyroptosis can affect the development of tumors. The relationship between pyroptosis and tumors is diverse in different tissues and genetic backgrounds. In this review, we provide basic knowledge of pyroptosis, explain the relationship between pyroptosis and tumors, and focus on the significance of pyroptosis in tumor treatment. In addition, we further summarize the possibility of pyroptosis as a potential tumor treatment strategy and describe the side effects of radiotherapy and chemotherapy caused by pyroptosis. In brief, pyroptosis is a double-edged sword for tumors. The rational use of this dual effect will help us further explore the formation and development of tumors, and provide ideas for patients to develop new drugs based on pyroptosis. Abstract Currently, pyroptosis has received more and more attention because of its association with innate immunity and disease. Publication types Research Support, Non-U. Gov't Review.

Under the category of necrotic cell death, there are many well-characterized, as well as some newly described, processes of cellular destruction. Contrary to classical thinking, necrotic cell death is not always physical and accidental in nature. Cells may trigger specific programmed self-destruct pathways when they are under stress and other methods of cell death are not feasible. Pyroptosis is a type of programmed necrotic cell death that is activated upon intracellular infections from bacteria, viruses, fungi, and protozoa in the presence of pathogen-associated molecular patterns PAMPs or cell-derived damage-associated molecular patterns DAMPs. It is typically induced in cells of the innate immune system, such as monocytes, macrophages, and dendritic cells.
Journal of Neuroinflammation volume 19 , Article number: Cite this article. Metrics details. It differs from other types of programmed cell death such as apoptosis, autophagy, necroptosis, ferroptosis, and NETosis in terms of its morphology and mechanism. As a recently discovered form of cell death, pyroptosis has been demonstrated to be involved in the progression of multiple diseases. Recent studies have also suggested that pyroptosis is linked to various ocular diseases. In this review, we systematically summarized and discussed recent scientific discoveries of the involvement of pyroptosis in common ocular diseases, including diabetic retinopathy, age-related macular degeneration, AIDS-related human cytomegalovirus retinitis, glaucoma, dry eye disease, keratitis, uveitis, and cataract. We also organized new and emerging evidence suggesting that pyroptosis signaling pathways may be potential therapeutic targets in ocular diseases, hoping to provide a summary of overall intervention strategies and relevant multi-dimensional evaluations for various ocular diseases, as well as offer valuable ideas for further research and development from the perspective of pyroptosis. Programmed cell death PCD plays an important role in regulating stress response, homeostasis, and diseases.

In-depth study of ASC and its upstream and downstream molecules or pathways may deepen our comprehension of the molecular mechanism of melanoma and provide new molecular targets for the design of anti-melanoma drugs. We discuss the attribution of pyroptosis in reprogramming tumor microenvironments and restoration of anti-tumor immunity and its potential application in cancer immune therapy. Engagement of the PD-1 immunoinhibitory receptor by a novel B7 family member leads to negative regulation of lymphocyte activation. Conventional cancer treatments such as chemotherapy and radiotherapy tend to kill cells that are in a rapidly proliferating status, including rapidly growing cancer cells and normal cells e. Biomed Pharmacother. PD-L1-mediated gasdermin C expression switches apoptosis to pyroptosis in cancer cells and facilitates tumour necrosis. In non-canonical inflammasome pathway, cleavage of GSDMD at Asp is executed by active murine caspase or its human homologs caspase-4 and -5 upon direct binding of cytosolic LPS [ 21 , 33 , 34 , 35 ] Fig. Sun, R. Pigment Cell Melanoma Res. The gasdermin proteins do not need to kill all cancer cells but activate tumor immunity by killing a small number of cancer cells. Absolutely with you it agree. In it something is also to me it seems it is excellent thought. Completely with you I will agree.
It is similar to it.
And it is effective?
