Nathaniel Essex was born in Victorian England during the 19th Century. He was obsessed with Charles Darwin's Theory of Evolution. Although he was genuinely passionate about his work, he also cared a great deal for his wife Rebecca Essex and their son. Disaster struck when their son died, aged four, of various birth defects. Essex started to intensify his work and theories about evolution to cope with the pain of this loss, ignoring his again pregnant wife as a result. Where he was once a passionate and tender man, he became more and more secluded and ignorant to the people around him. Unbeknownst to his wife, Essex even utilized his son's body as a test subject in hopes of figuring out what had caused his death.

Created by writer Chris Claremont , the character was first mentioned as the employer behind the team of assassins known as the Marauders in The Uncanny X-Men December , and later seen in silhouette in The Uncanny X-Men , with both issues serving as chapters of the " Mutant Massacre " crossover. His appearance was designed by artist Marc Silvestri. A villain who usually prefers to act through agents and manipulation, Mr. Sinister is born Nathaniel Essex in Victorian London. A human scientist, Essex is inspired by the work of his contemporary Charles Darwin and becomes obsessed with engineering humanity into a perfect race of superhumans. As he learns about mutants superhuman beings born with the X-gene , Essex encounters the mutant villain Apocalypse. The two become allies and Apocalypse uses alien Celestial technology to transform the British scientist into Mr. Sinister, an ageless man with super-powers. In the modern-day, Sinister develops a great interest and protective attitude towards the mutant heroes Cyclops and Jean Grey , believing their DNA can create the ultimate mutant. This and other factors lead him to have repeated clashes with the X-Men a group Cyclops and Jean Grey helped found and related teams. Several co-existing Mr. Sinister clones all sharing the original's memories and basic personality later formed a community. This community was later destroyed, but one clone that was born with an X-gene making him a mutant survived and continues to operate to this day. Sinister is often associated with the characters Apocalypse, Cable , and Madelyne Pryor , and is leader of the team of trackers and assassins known as the Marauders.

Nathaniel Essex, a brilliant scientist and geneticist, dedicating his his life's work to create the perfect mutant. Because of his work and of his mutant powers, Sinister was the first Apocalypse brought in his fold, and the first Horsemen as well. Sinister was at Apocalypse's side when they witnessed the battle that killed Charles Xavier. It was the same day Apocalypse declared his war on humankind. During these early days of his partnership with Apocalypse, Sinister adopted two brothers with exceptional mutant abilities. The Summers brothers, Alex and Scott , were the closest Essex had found to genetic perfection. Scott's genes were of particular interest to Sinister; he hoped to one day use them to create the most powerful mutant on the planet.

Through the years, Mister Sinister became one of the greatest geneticists to have ever lived, skilled in cloning , creating new life, and even in the manipulation of mutant DNA for enhancing powers, incorporating mutant DNA to his own. Mister Sinister spent most of his life scheming ways to break free from his designer, Apocalypse. When Professor X established the mutant island nation of Krakoa , Mister Sinister was granted not only citizenship, but also a seat on the Quiet Council of Krakoa , the Krakoan supreme government. Mister Sinister is a clone of the original Nathaniel Essex , one of three created in [48] along with a clone of Essex's late wife Rebecca [49] to explore different solutions to the problem of inevitable Machine supremacy. Like the rest of of Nathaniel's clones, he believed himself to be the only one and the original Nathaniel Essex, having no memory of his death in Bedlam. In , Mister Sinister awoke Apocalypse from his slumber once again, only to betray his master by infecting him with a techno-organic virus he had bred at Apocalypse's behest.

Logan, however, did not return unscratched, since it was here that Logan's memories were wiped clean. Mister Sinister has the ability to shape-shift from a humanoid form to an amorphous one. They have a son, Nathan Summers. Besides this area, he is also a skilled doctor, one of the best from his time in England with honors in medicine and later in biology. Now bitter and increasingly desperate regarding her missing child, Maddie believes Scott completely abandoned them and never cared enough to contact her or look for her. Sabretooth is enlisted as a leader in Sinister's new group of Marauders. After the " Ultimatum Wave ", he reforms his body and gets a job at Roxxon as part of their " brain trust ". At the same time, Sinister created a group of superpowered beings called The Children, which he was forced to give up to John Sublime, after Sabretooth killed the first of the Children: Hans. Mister Sinister was an Apocalypse-genetically created mutant hellbent on creating the perfect X-gene clone. Nathaniel Essex was born at Milbury House on the outskirts of London.

Sinister continues his science experiments and creates mutant chimeras to increase his army. His intellect later grew to new levels after his time with Apocalypse and the advanced Celestial technology he had access to, mastering not only this and all technology from Earth allowing him considerable expertise in cybernetics, robotics and computers but also new sciences to aid him in his goals to create the perfect mutant. After witnessing the death of a test subject that is a copy of Cyclops, Sinister is defeated by the X-Men and taken to the authorities. Sinister names the genetically engineered boy " Nathan Grey ", deciding that while Jean Grey was the boy's mother, he himself was the father. As her mind and health deteriorate, she becomes a hospital patient in San Diego for the rest of her life, her treatment provided by Sinister himself, who visits annually in his human guise of Nathan Essex. Discovering the house is completely empty, he concludes Maddie left with Nathan, deliberately leaving no trace of where she might be. Essex was in great turmoil, wanting to be a good man, but also not wanting the restrictions for his research that he had with his human morality. Seeing Maddie's resemblance to Jean and hearing about the timing of her experience in a plane crash, Scott wonders if Pryor is somehow his first love reborn. With her increased power, Jean creates a new body for her consciousness and the cosmic Phoenix Force to occupy, while creating a healing cocoon to repair the damage done to her original body. Sinister sets up a lab in Saigon and has his agent Scalphunter bring him soldiers and civilians for experimentation. Eventually the two wed and produced a child, Nathan Christopher Charles Summers.
