Dead Space: Downfall is the animated horror prequel to Dead Space , taking place during the events of Dead Space: Extraction and the Dead Space comic leading all the way up to the beginning of the first game. A screen crackles to life as an exhausted Alissa Vincent gets in the chair in front of the recording device, explaining the fall of the Ishimura due to an infestation of parasitic alien-organisms brought about by an artifact discovered on the surface of Aegis VII and tells the recipient of the message that the Ishimura and the artifact must be destroyed. The message ends. Going back in time, three patrolling inspectors in RIGs move to inspect unusual seismic activity on the planet surface. One of them notes chemical weathering on the walls and another takes note of what appears to be a claw mark. They discover an object referred to as a " Marker " on the planet's surface; the artifact is later brought aboard the Ishimura by orders of Captain Mathius.

Chronologically set prior to the events of the original Dead Space and after the Image Comics series, Dead Space: Downfall follows the events that take place aboard the Ishimura space station just before Isaac Clarke and his crew come onboard. The Ishimura is carrying a monolith called the Marker from the planet Aegis 7, and as things go in science fiction horror stories, this doesn't go as planned. The Marker has strange powers and begins to biologically alter and infect the many people onboard, turning them into freakish creatures known as Necromorphs.

Necromorphs are mutated corpses reshaped into horrific new forms by a recombinant extraterrestrial infection derived from a genetic code etched into and transmitted by the Markers. The resulting creatures are extremely aggressive and will attack any living organism on sight. The sole purpose of all Necromorphs is to acquire more corpses to convert and spread the infection. They are believed to be the heralds of humanity's ascension by certain Unitologists and individuals suffering from Marker Dementia , but on a more practical level, they are the extremely dangerous result of the exposure of dead tissue to the Markers ' signal. The Necromorphs are controlled via the Markers by a group of entities known as the Brethren Moons : massive, highly intelligent beings created after a Convergence Event is initiated by a Marker. The Necromorphs' sole drive is to create a mass of necrotic flesh that can be collected by the Markers during Convergence with the intention of creating another Brethren Moon, perpetuating the Moons' galaxy-wide cycle of extinction. The generation of a Necromorph begins with the Markers. These structures broadcast a highly concentrated electromagnetic signal that can alter dead tissue in range on a cellular level, converting it into pure Necromorph tissue and re-animating the corpse of the host organism. The infection is also spread directly through the Necromorph pathogen — that is, the reanimated cells created by the Markers and which compose the Necromorphs. Typically, the infection is passed on to new host bodies via specialized Necromorphs such as Infectors or Swarm Infectors.
